Kyprolis is indicated for the treatment of relapsed multiple myeloma in adult patients as part of combination therapy with lenalidomide and dexamethasone or with dexamethasone alone who have received at least one prior line of therapy.

Kyprolis is administered at an initial dose of 20 mg/m² (maximum dose 44 mg) on ??days 1 and 2 of cycle 1. If this dose is tolerated, the dose should be increased to 27 mg/m² on day 8 of cycle 1 (maximum dose 60 mg). Beginning with cycle 13, Kyprolis doses on days 8 and 9 are omitted.
